Project description
Third-cycle subject: Biotechnology
The focus of the research performed at the department is to develop predictable and robust systems for protein production, purification and detection. Well-characterized, small and folded domains descending from the bacterial receptors staphylococcal protein A (SPA) or streptococcal protein G (SPG) are used as scaffolds. Their known three-dimensional structures, a capability of independent folding and the absence of disulfide bridges make these domains ideal as frameworks for further protein engineering. Protein-engineering strategies are used in order to custom-make proteins for protein purification processes. The designed purification tags and antibodies produced within the HPA program are combined to develop strategies for capture of proteins and protein complexes in an efficient and selective manner. Also binding proteins for use in cancer diagnostics as well as therapeutics have been designed and are now optimized to achieve high selectivity and specificity in vivo. Furthermore, high throughput methods for production and purification of proteins are developed. Depending on protein and aim we are using either mammalian cells or bacteria for the protein production. (https://www.kth.se/profile/sophia/page/the-hober-lab)
We now look for a highly motivated doctoral student, to be engaged in an ongoing project. Within this project we aim to develop an affinity protein-platform for selection and characterization of affinity proteins. The new binders will be utilized for different purposes. The first aim is to develop robust, effective and efficient protein purification methods for different important target molecules. The other aim is to develop therapeutic protein domains that can be tailored to specifically bind disease-causing proteins.
Supervision: The doctoral student will be supervised by: professor Sophia Hober
