School of Engineering Sciences at KTH

Job description

Sugar transporters (GLUTs) have the fundamental role to take up energy into cells in the form of glucose and other hexoses; they belong to the MFS superfamily and follow a rocker-switch alternating access model of transport. Due to their ubiquitous nature and their overexpression in cancer cells, GLUTs are important targets for drug design. Yet, while multiple structures have been resolved, their dynamic nature and the distinct relative stability of different states in related isoforms only allow for a poor resolution of the different steps along the transport cycle.
Recent work on bacterial sugar transporters has revealed an imperfect proton coupling in proteins previously believed to be passive transporters, suggesting that the destabilization of intermediate states is at the core of the substrate:proton coupling. The aims of this project are to characterize the proton-coupling mechanism affecting GLUTs dynamics, its and allosteric regulation by small molecule inhibition.
The postdoc candidate will carry out computational work, using mainly molecular dynamics simulations, including but not limited to enhanced sampling simulations and constant pH. Results from this work will be tested by our collaborators in the Drew lab at Stockholm University.
